About Composition

Azithromycin:

Azithromycin is a broad-spectrum macrolide antibiotic. It is used in the treatment of the respiratory tract infection, genitourinary tract infection.

Fluconazole:

Fluconazole is the antifungal drug. It is used to treat systemic and superficial fungal infections. It is mainly efficacious in the treatment of vaginal yeast infection

Secnidazole:

Secnidazole is the 5-nitroimdazole antimicrobial agent. It is effective against anaerobic gram positive and gram-negative bacteria

Mechanism of action

Azithromycin inhibits the protein synthesis by binding with the 23S rRNA of the 50 s ribosomal unit of the RNA and inhibit the translocation step in the protein synthesis.

Fluconazole is inhibiting the enzyme lanosterol 14-alpha demethylase which is responsible for the conversion of the lanosterol to ergosterol which is the essential component for the cell wall synthesis of fungi

Secnidazole is a prodrug. It is converted to an active metabolite by reducing the nitro group into anion by bacterial enzymes which then interfere with the DNA synthesis in the bacteria

Pharmacokinetics

Azithromycin:

Absorption: It is orally absorbed.
Protein binding: It is bound to proteins to lesser extend.
Metabolism: It is metabolized in the liver.
Excretion: It is excreted via urine.

Fluconazole:

Absorption: It is absorbed orally and produce bioavailability up to 90%.
Protein binding: protein binding of fluconazole is low.
Metabolism: Fluconazole is undergoing glucuronide conjugation.
Excretion: It is excreted unchanged in the urine

Secnidazole:

Absorption: It is completely absorbed by oral route.
Protein binding: protein binding is 5-15%.
Metabolism: It is metabolised by oxidation reaction.
Excretion: It is excreted unchanged in the urine

Side Effects

  • Headache
  • Nausea
  • Abdominal pain
  • Itching or discomfort in the vagina

Contraindications

  • Contraindicated in the people with known hypersensitivity.
  • Contraindicated during the third trimester of pregnancy
